The ALK/PAX3 FISH Probe Kit is designed to detect the human ALK and PAX3 gene regions located on chromosome bands 2p23.2 and 2q36.1, respectively. Rearrangements and abnormal expression of the ALK gene region – also known as CD246 or NBLST3 – have been observed in NSCLC, neuroblastoma, inflammatory breast cancer, rhabdomyosarcoma, anaplastic large cell lymphoma and other malignancies. ALK amplification can also serve as a mechanism of acquired resistance to ALK TKI therapy in NSCLC. Rearrangements and abnormal expression of the PAX3 gene – also known as WS1, WS3, CDHS or HUP2 – have been observed in alveolar rhabdomyosarcoma (ARMS) and other tumor types and conditions.

The probe is labeled with CytoOrange, CytoGreen and CytoAqua. CytoOrange is a fluorophore with an excitation peak at 551 nm and emission peak at 575 nm, giving it an orange fluorescent signal with an appropriate filter; CytoGreen has an excitation peak at 495 nm and emission peak at 518 nm, giving it a green fluorescent signal with an appropriate filter; CytoAqua has an excitation peak at 422 nm and emission peak at 471 nm, giving it an aqua fluorescent signal with an appropriate filter.
